Rationale for Study
This exploratory study was conducted to provide in-vivo effectiveness and safety data for a cosmetic whitening tooth polish. The data from this study was used to validate and substantiate the following claims:
- For whiter teeth
- Clinically tested
- Dentist approved (in relation to the safety of the product only).
Study objectives
Primary Objective
The primary objective was to evaluate the efficacy of the tooth polish in whitening the colour of teeth when using the Vita 3d master shade guide at baseline, Day 15 and Day 29 to measure changes in colour.
Secondary Objective
The secondary objective was to evaluate the efficacy of the tooth polish in whitening the colour of teeth when capturing images and assessing L*a*b measurements of the teeth at baseline, Day 15 and Day 29 to measure changes in colour.
Tertiary Objective
The tertiary objective was to evaluate the safety of the product with oral health checks of the soft and hard tissue of the mouth by a dentist at baseline, Day 15 and Day 29.
Study Summary
Study Protocol
The study was performed in accordance with Alba Science Ltd. study protocol 212737 (final version 1.4 dated 16 March 2017).
A copy of the study protocol is included in Appendix A.
Study Description
This was a single-centre, within-subject study performed in a population of 34 subjects testing a whitening tooth polish.
Recruited subjects attended for screening assessments having refrained from eating, drinking (except for water) or any oral hygiene for 1 hour prior to their screening appointment. Each subject signed a consent form confirming their agreement to take part in the study. They were then screened for specific inclusion/exclusion criteria by trained personnel and assessed for oral parameters by a dentist, including an oral health check (soft and hard tissues) and a baseline Vita shade score.
Once eligibility was confirmed, enrolled subjects had baseline images taken of their teeth using Alba Science standard image capturing equipment. Subjects wore dental retractors and the images were captured in a room where all external light sources were removed. Subjects were dispensed their allocated test product, along with a standard toothpaste and toothbrush, diary and instructions for use. Subjects were instructed to start using the standard toothpaste twice daily and test product once a day for the next 28 days. They were instructed not to use any other toothpaste or mouthwash products.
Subjects attended for further visits at the test centre on Days 15 and 29, having refrained from eating, drinking (except for water) or any oral hygiene for 1 hour prior to each visit. At these visits, subjects had further oral health checks performed, Vita shade scoring and image capture.
Primary Endpoint
The primary endpoint was the change in Vita shade scores from baseline to Day 15 and Day 29 to measure changes in colour.
Secondary Endpoint
The secondary endpoint was the change in L*a*b measurements of the teeth at baseline to Day 15 and Day 29 to measure changes in colour.
Tertiary Endpoint
The tertiary endpoint was any findings from the oral health checks performed of the soft and hard tissue of the mouth by a dentist at baseline to Day 15 and Day 29.
Discussion And Conclusion
The primary study objective was to evaluate the efficacy of a tooth polish in whitening the colour of teeth when using the Vita 3d master shade guide at baseline, Day 15 and Day 29 to measure changes in colour.
Statistical analysis of the Vita 3d Master Shade data from 33 evaluable subjects showed that once daily use of Laviish Tooth Polish, with twice daily brushing using a standard tooth paste, resulted in a significant change in tooth shade (lightening of between 4 and 5 shades) after 2 weeks and 4 weeks use, when compared with baseline.
The secondary study objective was to evaluate the efficacy of the tooth polish in whitening the colour of teeth when capturing images and assessing L*a*b measurements of the teeth at baseline, Day 15 and Day 29 to measure changes in colour.
Statistical analysis of the L*a*b* measurements showed that there was a significant decrease in b* at 4 weeks (i.e. a colour change towards the blue chroma), after once daily use of Laviish tooth polish, with twice daily brushing using a standard toothpaste, indicating a whitening effect. There were no significant differences in b* at Day 15 and no significant differences from baseline in L* or a* measures at Day 15 or Day 29.
The tertiary study objective was to evaluate the safety of the product with oral health checks of the soft and hard tissue of the mouth by a dentist at baseline, Day 15 and Day 29.
There were no changes in the oral health check assessments conducted by the study dentist at any time point post-baseline.

Use once a day after brushing your teeth with a regular toothpaste. Dip a separate clean dry toothbrush into the polish and coat generously. Brush gently in a circular motion for 2 minutes. After use rinse mouth and toothbrush well with cold water. Remove any excess water and ensure the toothbrush is dry prior to next application.
